T4 DNA ligase는 duplex DNA 또는 RNA에 있는 5' phosphate and 3' hydroxyl 끝 말단 부위 사이 phosphodiester bond 형성을 촉진한다.
Ligase는 duplex DNA , RNA, 또는 DNA/RNA hybrids 에 있는 단일가닥 틈 사이를 repair 할 뿐만 아니라 blunt-end 그리고cohesive-end 끝말 단 부위로부터 DNA를 ligate 시킨다.
T4 DNA Ligase는 재조합 E.coli strain 분리하였다.
