Wide linear dynamic range & high PCR efficiency

  • 10 ~ 1010 copies의 광범위한 증폭 범위
  • MIQE guide line을 준수하는 90 ~ 110%의 qPCR 증폭 효율

qPCR template로 purified DNA를 10배씩 serial dilution 하여 사용하였으며, 2번 반복 실험되었습니다.

qPCR Service는 MIQE 가이드라인을 따르고 있으며, 숙련된 전문가들에 의해 SCI 논문에 그대로 활용 가능한 정확하고 신뢰성 있는 분석 결과를 빠른 기간 내에 제공해 드립니다.

  • Step 1 : RNA Qualitiy Check
    의뢰된 RNA 샘플은 qPCR에 적합한지 판단하기 위해 Caliper사의 LabChip™GX를 이용하여 RNA Quality Index (RQI)를 측정합니다. RQI를 통과하였을 경우 다음 과정을 진행하며, 적합하지 않을 경우 고객과 상담 후 다음 단계를 진행합니다. 
  • Step 2 : cDNA Synthesis (Reverse transcription)
    분석에 적합한 RNA 샘플은 cDNA로 합성하게 됩니다. 고온에서도 안정적인 합성효율을 보이는 Accupower® Rocketscript™ Cycle RT Premix with oligo(dT)20를 이용하여 cDNA 합성을 진행합니다. 
  • Step 3 : qPCR
    cDNA는 Accupower® 2X GreenStar ™ qPCR Master Mix와 검증된 타겟 특이 primer가 포함되어 있는 Exicycler™ 96 Real-Time Quantitative Thermal Block 을 이용하여 qPCR을 진행합니다. 
  • Step 4 : Data Analysis 
    qPCR data는 자체 개발한 분석 소프트웨어로 분석한 후 엑셀로 변환하여 제공하며, raw Ct 값, melting curve analysis data, p-value, fold change, scatter plots, volcano plots, heatmap image 등도 분석결과에 포함됩니다.
